Established in October 2023, the A*STAR SIgN-BCA Centre of Excellence (CoE), the Single Cell Cloning Platform, utilizes the Beacon Optofluidic System, an innovative technology that seamlessly combine Optoelectronic Positioning (OEP) and microfluidics to offer rapid and precise cell sorting, single-cell functional analysis, and real-time monitoring of cellular interactions and responses. Highly efficient and high-throughput, the platform facilitates advanced single-cell research and supports biopharmaceutical applications.
The objectives of the COE are to:
- Promote as a regional hub for scientific innovation, training, and research services using the Beacon system.
- Provide A*STAR and Singapore’s research community with access to Bruker Cellular Analysis’s cutting-edge technology for antibody discovery, immune cell profiling, and cell line development (Research use only).
- Accelerate innovation by establishing co-development projects for customized single-cell applications.

Cell Line Development
The Edinburgh Genome Foundry (EGF) is a research facility specialised in the modular assembly of DNA constructs using a highly automated robotic platform. Edinburgh Genome Foundry have the first Beacon instrument installed in an academic institution in Europe. This high-throughput automated cell selection platform allows high-capacity single cell cloning, analysis and outgrowth, with FDA-approved verified clonal origin. EGF is able to run campaigns for Cell Line Development and Antibody Discovery with the Cell Therapy workflow to follow soon.

Monash Antibody Discovery Platform (MADP) is a world-class antibody platform servicing Monash University and the broader scientific community.
MADP provides services for antibody discovery, antibody production, antibody sequencing, flow cytometry analysis and cell line generation. The primary focus in the core is generating high-affinity monoclonal antibodies that are specific for the customer’s needs, but we also offer a range of additional services related to cell line work and antibody characterisation.
The Beacon® Optofluidic System located in MADP is configured for antibody discovery. Using the Opto™ Plasma B discovery workflow, single cells are loaded into wells of an Optoselect chip and then assayed for hits. Cells that give a positive signal are either exported into a 96-well plate for mRNA extraction or have mRNA capture beads added and undergo on-chip cell lysis. From here cDNA is generated by reverse transcription and heavy and light chains are sequenced. This whole process takes only days.
