Linking single-cell spatial multiomics of TCR repertoires using CosMx SMI with live T-cell function using the Beacon to characterize immune cell dynamics in tumors

This poster demonstrates how the CosMx® Spatial Molecular Imager (SMI) combines spatial TCR repertoire profiling, whole-transcriptome analysis, and high-plex protein measurements to identify tumor-reactive T cells within their native tumor microenvironment. By linking functional live T-cell data from the Beacon® optofluidic platform with spatial multiomics, the approach reveals mechanisms of immune activation, exhaustion, and immune evasion that may inform biomarker discovery and patient-specific immunotherapy strategies.

Spatial TCR profiling at single-cell resolution: CosMx SMI detects TCR segments and clonotypes alongside whole-transcriptome and protein data in FFPE tumor samples, enabling detailed mapping of T-cell populations and their spatial context.
Linking function to location: T cells identified on the Beacon platform as capable of killing tumor cells were traced back into tissue sections, revealing where tumor-reactive, exhausted, and bystander T cells reside within tumors.
Understanding immune evasion: The combined datasets show how factors such as T-cell exhaustion, regulatory T cells, extracellular matrix barriers, and local tumor microenvironments may limit anti-tumor immune responses despite the presence of infiltrating T cells.
