Function-Driven Single-Cell Screening and Characterization of Bispecific T Cell Engagers (BiTEs)
Bispecific T cell engagers (BiTEs) redirect T cells to tumor cells by simultaneously binding CD3 and tumor-associated antigens, enabling targeted and immune-mediated killing. However, candidate ranking is typically based on bulk cytotoxicity or cytokine assays, which mask underlying functional heterogeneity across individual T cells. Single-cell functional approaches resolve these challenges by enabling more informed, confident, and efficient candidate selection.
